My research focuses on biomonitoring of carcinogen-DNA adducts in humans derived from environmental and endogenous genotoxicants. Employing advanced techniques of LC-mass spectrometry, I established methods to measure DNA adducts of aristolochic acids (AA), a renal carcinogen found in herbal plant Aristolochia, at a trace level employing freshly frozen and formalin-fixed paraffin embedded (FFPE) human tissues. The FFPE tissue block is the most common method to preserve tissues obtained from biopsy, however, it has not been utilized for DNA biomarker discovery due to technical difficulties. Our methods of DNA retrieval from FFPE tissues can fully reverse the crosslinks in the tissues under a mild condition and preserve the DNA adducts during the tissue process. The development of the DNA retrieval method for FFPE tissue is a major technological advancement in DNA adduct biomarker research. Recently, the method of DNA retrieval from FFPE has been extended for DNA adducts of multiple classes of carcinogens including aristolochic acids (AA), heterocyclic aromatic amine (HAAs), aromatic amine, polycyclic aromatic hydrocarbons (PAHs), and tobacco carcinogens. The methodology is now primed to examine various adducts in human cohorts where fresh frozen tissue samples are not available.
